In vivo comparison of the HUFA biosynthesis capacities between the Eurasian perch and the rainbow trout

The main purpose of this study is to estimate the potential of the perch to synthesize HUFA, and to compare this potential with that of the trout. To this end, perch and trout will be fed for at least 4 months with a control diet formulated with fish ingredients, or an experimental diet completely deprived in HUFA formulated with plant sources.
During the nutritional challenge, the food intake for each diet and each fish species will be calculated, and parameters of growth will be measured (length and weight).

At the end of the experiment, transcriptomic and enzymatic measures will be investigated on samples of liver and intestines. This approach will be completed by the modelling of the fatty acids metabolism based on the complete physical balance assessments according to the Turchini method. These measures focused on the fatty acids metabolism will be completed by physiological measures of immune parameters.

These approaches dedicated on certain molecular actors will be completed by a global approach on the proteome. This last one, based on the 2D-DIGE approach, will allow riddling the proteome in target organs (such as the liver) in response to a modification of diet composition (with or without plant sources). This comparison by proteomic will allow to characterize the protein modulation induced by the use of a diet deprived in HUFA.
